ADAM 10 is responsible for Her 2 shedding. Truncated Her 2 receptor has enhanced activity.
Trastuzumab inhibits this shedding.
SHEDDASES
Sheddases are membrane bound
enzymes, usually members of the
ADAM ("a disintegrin and metalloprotease")
family, that cut off, shed, extracellular
portions of transmembrane proteins

Hence sheddases demultiply the yield of agonists
Sheddase inhibitor,
